Dual transcriptomics of virus-host interactions: comparing two Pacific oyster families presenting contrasted susceptibility to ostreid herpesvirus 1 ArchiMer
Segarra, Amelie; Mauduit, Florian; Faury, Nicole; Trancart, Suzanne; Degremont, Lionel; Tourbiez, Delphine; Haffner, Philippe; Barbosa-solomieu, Valerie; Pepin, Jean-francois; Travers, Marie-agnes; Renault, Tristan.
Background: Massive mortality outbreaks affecting Pacific oyster (Crassostrea gigas) spat in various countries have been associated with the detection of a herpesvirus called ostreid herpesvirus type 1 (OsHV-1). However, few studies have been performed to understand and follow viral gene expression, as it has been done in vertebrate herpesviruses. In this work, experimental infection trials of C. gigas spat with OsHV-1 were conducted in order to test the susceptibility of several bi-parental oyster families to this virus and to analyze host-pathogen interactions using in vivo transcriptomic approaches. Results: The divergent response of these oyster families in terms of mortality confirmed that susceptibility to OsHV-1 infection has a significant...

Real-time PCR assay for rapid detection and quantification of Vibrio aestuarianus in oyster and seawater: A useful tool for epidemiologic studies ArchiMer
Saulnier, Denis; De Decker, Sophie; Haffner, Philippe.
Because Vibrio aestuarianus is known to cause serious infections in Pacific oyster Crassostrea gigas, a real-time PCR assay was developed targeting the dnaJ gene of this bacterium. Only V. aestuarianus strains isolated from C. gigas mortality events in different geographic areas and the reference strain tested positive, whereas no amplification products was obtained with type strains belonging to 23 other species of Vibrio. Sensitivity and reproducibility of the method were assessed using either seawater or oyster homogenate samples spiked with one V aestuarianus strain. All these samples were stored at -20 degrees C in order to mimic retrospective or grouped natural sample analysis without quantification bias due to prolonged freezing. Analysis of...

A field report on vibrio disease of seabass (Dicentrarchus labrax ) in the South of France ArchiMer
Breuil, Gilles; Haffner, Philippe.
Vibrio anguillarum was isolated from diseased seabass reared in aquatic farms (hatcheries and grow-out farms) located along the French Mediterranean coast. A representative strain (V62) differs from the French V. anguillarum strain (V408) by its ability to grow on a 0% salinity medium. The pathogenicity of the representative strain for sea bass was confirmed by injections and bath challenge. The V62 strain reacted with V. anguillarum antiserum (V408) and 84% of the isolated strains from diseased sea bass agglutinated with anti V62 serum.

Species-specific mechanisms of cytotoxicity toward immune cells determine the successful outcome of Vibrio infections ArchiMer
Rubio, Tristan; Oyanedel, Daniel; Labreuche, Yannick; Toulza, Eve; Luo, Xing; Bruto, Maxime; Chaparro, Cristian; Torres, Marta; De Lorgeril, Julien; Haffner, Philippe; Vidal-dupiol, Jeremie; Lagorce, Arnaud; Petton, Bruno; Mitta, Guillaume; Jacq, Annick; Le Roux, Frederique; Charriere, Guillaume; Destoumieux-garzon, Delphine.
Vibrio species cause infectious diseases in humans and animals, but they can also live as commensals within their host tissues. How Vibrio subverts the host defenses to mount a successful infection remains poorly understood, and this knowledge is critical for predicting and managing disease. Here, we have investigated the cellular and molecular mechanisms underpinning infection and colonization of 2 virulent Vibrio species in an ecologically relevant host model, oyster, to study interactions with marine Vibrio species. All Vibrio strains were recognized by the immune system, but only nonvirulent strains were controlled. We showed that virulent strains were cytotoxic to hemocytes, oyster immune cells. By analyzing host and bacterial transcriptional...

Abnormal mortality outbreaks occurred in the majority of Pacific oyster (Crassostrea gigas) producing areas in France in 2008 during the summer period. These outbreaks were usually sudden and severe (up to 100%) and affected essentially spat (oysters less than 1 year-old) and juveniles (12 to 18 month-old oysters). Crassostrea gigas was the single affected species Mortality outbreaks were first observed from mid May to mid June in Brittany, Vendée, Charente Maritime and Mediterranean coast A second episode of massive mortality events was reported at the end of June and the beginning of July. All oyster production sites were affected except some particular locations including Arcachon Bay and Brittany. Mortality occurred latter in Arcachon Bay in August....

Mass mortalities in hatchery-reared sea bass (Lates calcarifer) larvae associated with the presence in the brain and retina of virus-like particles ArchiMer
Renault, Tristan; Haffner, Philippe; Baudin Laurencien, F.; Breuil, Gilles; Bonami, Jean-robert.
Since 1986, an unknown disease causing high mortalities appeared in Ifremer's Pacific OceanologicaI Center (Tahiti) nursery of sea bass (Lates calcarifer). opathology of lesions involving both centraI nervous system (CNS) and eye retina, and the tuation exists in South-East Asian countries (Indonesia, Philippines, Malaysia and Singapore), and in northem Australia. This paper describes the histmorphology of a new fish virus, which is believed to be the aetiological agent of high mortalities among sea bass larvae.

Ecologically realistic model of infection for exploring the host damage caused by Vibrio aestuarianus ArchiMer
Parizadeh, Leila; Tourbiez, Delphine; Garcia, Celine; Haffner, Philippe; Degremont, Lionel; Le Roux, Frederique; Travers, Marie-agnes.
Although vibrios are frequently associated with marine organisms mortality outbreaks, knowledge on their ecology and pathogenicity is sparse, thus limiting disease management and prophylactic strategies. Here, we investigated V. aestuarianus infection onset and progression in the wild, taking advantage of a “claire” pond: a semi‐closed system with limited seawater renewal, theoretically more adapted to disease transmission. We showed a positive association of the bacteria with oysters, which can constitute a reservoir for the bacteria in the winter. Moreover, passage through oysters was found to be necessary for experimental disease reproduction as vibrios shedding from diseased oysters has higher infectivity than from in vitro grown. We next developed an...

Oyster diseases caused by pathogenic vibrios pose a major challenge to the sustainability of oyster farming. In France since 2012 a disease affecting specifically adult oysters has been associated with the presence of Vibrio aestuarianus. Here, by combining genome comparison, phylogenetic analyses and high throughput infections of strains isolated before or during the recent outbreaks, we show that virulent strains cluster into two V. aestuarianus lineages independently of the sampling dates. The bacterial lethal dose was not different between strains isolated before or after 2012. Hence the emergence of a new highly virulent clonal strain is unlikely. Each lineage comprises nearly identical strains, the majority of them being virulent, suggesting that...

Development of an antiseptic procedure to improve retention rates during the grafting process of black-lip oyster, Pinctada margaritifera ArchiMer
Cochennec, Nathalie; Haffner, Philippe; Levy, Peva; Saulnier, Denis; Fougerouse, A.; Langy, Sandra.
In French Polynesia, black pearl cultivation now takes place on about 30 of the 118 atolls or islands. This activity has largely replaced copra, coconut oil and fishing as a source of income. Around 1,000 pearl farms, generally small ones of less than 5 ha, are registered. The pearl farms are mainly owned by families. In 1998, about 7,000 persons were employed in theentire pearl industry, from production to retailing. In value, this activity represents the first exportation of French Polynesian products. From 1988 to date, exports increased exponentially before leveling. Starting with less than two kilograms in 1972, exports have now stabilized at Il tons of "black" pearls.

Infectious diseases are mostly explored using reductionist approaches despite repeated evidence showing them to be strongly in fluenced by numerous interacting host and environmental factors. Many diseases with a complex aetiology therefore remain misunderstood. By developing a holistic approach to tackle the complexity of interactions, we decipher the complex intra-host interactions underlying Pacific oyster mortality syndrome affecting juveniles of Crassostrea gigas, the main oyster species exploited worldwide. Using experimental infections reproducing the natural route of infection and combining thorough molecular analyses of oyster families with contrasted susceptibilities, we demonstrate that the disease is caused by multiple infection with an initial...

Oyster Farming, Temperature, and Plankton Influence the Dynamics of Pathogenic Vibrios in the Thau Lagoon ArchiMer

Vibrio species have been associated with recurrent mass mortalities of juvenile oysters Crassostrea gigas threatening oyster farming worldwide. However, knowledge of the ecology of pathogens in affected oyster farming areas remains scarce. Specifically, there are no data regarding (i) the environmental reservoirs of Vibrio populations pathogenic to oysters, (ii) the environmental factors favoring their transmission, and (iii) the influence of oyster farming on the persistence of those pathogens. This knowledge gap limits our capacity to predict and mitigate disease occurrence. To address these issues, we monitored Vibrio species potentially pathogenic to C. gigas in 2013 and 2014 in the Thau Lagoon, a major oyster farming region in the coastal French...

Mass mortality outbreaks (90-100%) in both juvenile and adult blue mussels were reported in 2014 along the Pertuis Charentais area of the French Atlantic coast. A series of studies were carried out to learn more about the phenomenon through detecting pathogens, describing environmental conditions and establishing the physiological status of the animals. Bacteria identif ied as belonging to the V. splendidus species were detected in dying mussels and seem able to cause mortality in blue mussels in the laboratory. Unusual environmental conditions (large amounts of fresh water, re-suspension of sediments during a series of storms, and a high renewal rate of specific water masses in Brittany’s Pertuis area) may be partially responsible for sparking the phenomen

Since 1991, sporadic, but high mortality in larvae of the Pacific oyster (Crassostrea gigas) has been observed during the summer in various French hatcheries. This mortality was concomitant with the detection of a virus by transmission electron microscopy. This virus, by virtue of its morphological characteristics and developmental cycle, seems to belong to the Herpesviridae family. The pathogenicity of this viral agent has been demonstrated in the laboratory by experimentally induced mortality on healthy larvae. Moreover, concomitant mortality has been reported in larvae of Crassostrea gigas and Ostrea edulis reared in the same tanks, with detection of a herpes-type virus.
